Propriedades fisico-quimicas e interação com membranas biologicas dos surfatantes não-ionicos da serie Renex (nonilfenol etoxilado)

RESUMO

Concentration-dependent surfactant effects on erythrocite membranes are well-known, in the literature: high concentrations are haemolytic but low concentrations are often protective. These effects will depend on the pattern of surfactant interaction with the membrane, on surfactant-membrane binding and on its consequences, which in turn depend on surfactant structural characteristics: nature and sizes of polar and apolar groups as well as the presence (or not) of charges.This is a study on the effects of a well-known surfactant family, widely used in bilogical studies, on the erythrocite membrane. Surfactants used are ethoxylated nonylphenols, including two compounds with a high degree of ethoxylation. Renex brand (types 95, 200 and 1000 ) surfactants were used, with an average 9.5, 20 and 100 ethylene oxide residues per mol of phenol, respectively.Surfactant characterization was done by NMR and IR spectroscopies, which confirmed the stated composition and the average number of ethylene oxide residues per molecule.Two physico-chemical properties of Renex surfactants were determined, relevant for their interactions with membranes: i) critical micelle concentrations were determined by fluorimetry and by ESR; ii) spin-Iabel order parameter within micelles were determined by ESR, and give information regarding micelle apolar medium fluidity. Renex 95 has the lowest cmc (0.075 mM) and its micelles are the least fluíd. The cmc s of Renex 200 and 1000 are 0.11 and 0.75 mM respectively, and Renex 1000 micelle is the most fluid.Two other types of experiments were performed: i) surfactant effect on whole erythocite membrane within hyposmotic media was determined; ii) membrane ghost phospholipid solubilisation in surfactant solutions was evaluated. Renex 95 is haemolytic (at higher concentrations) and solubilizes phospholipids. On the other hand, Renex 1000 protects the cell against haemolysis and does not show the ability to solubilize the membrane
