Estrutura e ultra-estrutura da prostata ventral e vesicula seminal de camundongos isogenicos (C57B16/J) submetidos a ingestão cronica de alcool

RESUMO

Excessive alcohol consumption causes metabolic changes and pathologies in different systems of the organism Among them, the liver cirrhosis, alterations in the blood urine barrier, psychosis and infertility in male reproductive tract. It s related that alcohol affects directly the male gonads, altering the plasma testosterone levels, and indirectly, by injury in hypothalamic pituitary gonadal axis. However, little is know about the morphological alterations upon the organelles involved in the secretory process in the accessory sex glands due to long-term chronic ingestion of different alcohol dosages. The present investigation had for objective the evaluation of the macroscopic, microscopic and ultrastructural alterations provoked by the chronic alcohol ingestion of different ethanol rates during increasing periods of time upon the epithelial cells of ventral prostate and seminal vesicle of C57B16J mice. Sixty male adult mice were divided in three experimental groups: Control, alcoholic 25% and alcoholic 35% that received tap water and tap water containing ethanol diluted to 25° G.L and 35° G.L. respectively. AlI mice were fed with the same solid diet. After 150 and 250 days, the mice were sacrificed and the ventral prostate and the seminal vesicle were collected and processed for light and transmission electron microscopy. The structural alterations observed in the epithelial cells of the ventral prostate and the seminal vesicle of the alcoholic mice consisted of a reduction in epithelial size and in the cell volume, keeping the same proportion nuclear to cytoplasmatic ratio as observed in control groups. The major ultrastructural alterations observed in the alcoholic groups were: increased of area and density of area of eletrondense bodies in the basal layer of the cell, decreasing area and density of area of secretory vacuoles, dilated rough endoplasmatic reticulum and Golgi complex cistems and discontinuity of the microvilli. It was also observed the occurrence of irregularly shaped nuclei and condensed chromatin in the nuclei periphery and around the nucleoli. Apoptotic nuclei were observed in the alcoholic treated group 25% after 250 days. It s concluded that the chronic ethanol ingestion causes morphologic depletory alterations in the epithelial cells of the ventral prostate and the seminal vesicle negatively affect the secretory process of these glands
