Criopreservation of canine semen using associations of crioprotectors and two protocols to unfreeze / Criopreservação de sêmen canino, utilizando associações de crioprotetores e dois protocolos de descongelamento

RESUMO

The objective of this work was to evaluate the effect of permeable and unpermeable protocols in the same dilution media and to compare two unfrozen canine semen protocols. Three adults males from Labrador Retrievier breed, were submitted to weekly collection of semen for five weeks, in a total of 15 samples in each experimental design. The in vitro testes applied to the semen samples were evaluation of: the progressive motility, the spermatic vigor, the integrity of plasma and acrosome membrane and the sperm longevity. The base mean diluent utilized for the crioprotectors associations was tris-citrate: (G1) (modified tris-citrate = 3% dimetil-formamide + 3% glycerol); (G2) (modified tris-citrate = 3% dimetilformamide + threalose) and (G3) (tris-citrate = 4% glycerol). For the unfrozen semen, was used a fast unfrozen protocol (75 C/ 7) and the slow unfrozen protocol (37 C/ 1). A difference was detected (P<0.01) among the tested associations, and the G1 and G3 associations were superior to the G2 one. But, evaluating each protector association separately, there was no difference (P>0.01) when utilizing the fast and slow unfrozen protocols. These results showed that the glycerol cryopreserved sperm cell showed better results as compared to the others associations used in preserving the canine sperm cells.
