Proteinas cotiledonares de soja : detecção "in situ" e mobilização durante a germinação

RESUMO

Seeking to study the various changes in the protein content in seeds of Glycine max during germination, a series of general and cytochemical techniques was performed in order to study the protein bodies of this legume "in situ",verifing the presence of these bodies in the cotyledon cells by hematoxilin-eosin and xylidine-ponceau stains and silver impregnation. The quantification of protein was made by absorption measurements of the specimens stained with XP at various times and positions, indicating a mobilization of the reserve material of the cells far from the vascular system, to the interior of this system and then to the radicle of the embryo. The protein bodies were found to be membrane, evidenced by toluidine blue (TB), surrounded by a dueto the presence of basophilic material in this area. The cell walls of the cotyledon and radicle were stained by the TB,displaying metachromasy. During germination there was an increase in the metachromatic index of the stained substrate, indicating a decrease of negative radicaIs. In relation to the molecular organization there was a decrease in the cristalinity and agregation of the cell alls during germination along with a variation in the alycosaminoglycans content in the various areas analysed. The lectins present in the soybean seed are associated with the proteic material, thus losing their agglutinating properties in the ?in situ? material
