RESUMO

The oxidative degradation of fatty acids from soybean oil fraction, that occurs during the grain processing, develop a flavor of unripe been that modify the palatability and therefore the acceptability of soybean products, resulting in problems for the food industry. The enzyme lipoxygenases (linoleate: O2 oxidoreductase, EC 1.13.11.12), that catalyses this oxidation reaction, are present in soybean seed in the three isozyme forms encoded by three dominant genes, Lox1, Lox2 and Lox3. The selection assisted by molecular markers is an area that has great impact within breeding programs. Recently, a new class of molecular markers, called indel (insertion / deletion) and SNP (Single Nucleotide Polymorphism) have been used. These are based on detection of polymorphisms by the change of a single base in the genome. The identification of molecular markers within the genes encoding lipoxygenase may help in the selection of recessive alleles that determine the absence of lipoxygenase in the seed, accelerating the process of soybean breeding. The objective of this study was verify and identify the polymorphisms in the sequences of genes Lox2 and Lox3, respectively, between varieties of different genetic origin used in the Program of Improvement and quality of Soybean at BIOAGRO/UFV, and tovalidate some of these polymorphisms as molecular markers. A pair of primers for LOX2 and 14 pairs of primers designed to amplify the entire gene LOX3 were used to obtain genes fragments. The sequences were obtained from the direct sequencing of PCR products from each primer pair. Subsequently, a population of RILs contrasting for the presence / absence of seed lipoxygenases, were used for the validation of two SNPs as markers for genes Lox 2 and a indel marker for LOX 3. The genotyping method used was the direct sequencing of fragments that contained the polymorphisms studied. Two SNPs for LOX 2 and the indel contained in the first exon of the gene LOX 3 were co- segregated with the character presence/absence of LOX 2 and 3 respectively in the individuals of the population of RILs. These polymorphisms have the potential to be used as tools in soybean breeding assisted by molecular markers.
