Estudo do processo de produção de particulas lipidicas solidas e lipossomas : estudo de variaveis e encapsulação de medicamentos de primeiro tratamento da tuberculose

RESUMO

This work presents scale up studies for the production of so1id lipid particles (respirable powders) and liposomes, which are useful as vehic1es for contro1led delivery of rugs. The lipid particles were produced by spray drying, and the liposomes were prepared by hydration of the particles under high shear mixing. It was used a hydrogenated phospholipid, Epikuron 200 SH, as a structural component of the particles, and mannitol was used as excipient or core material. The tubercu1osis drugs Isoniazid and Pirazinamide were incorporated in both vehicles (powder and liquid formulation). The incorporation of excipients and the operational conditions influenced the process mass yield and the solid particles properties, enabling the modulation of the particles for specific app1ications. The lipid particles had a diameter of less than 5mm, and the efficiency of drug incorporation varied nom 20 to 1000Aa, depending on the drying operational conditions and the formulation. The incorporated drugs were gradually re1eased during 24 hours, behaving 1ike dried membranes. The liposomes properties were influenced by the impeller type and by the so1id particle microstructure. Mean diameters around 2000mn and with homogeneous size distribution were obtained using a high shear impeller produced by Kroma Equipamentos Especiais Ltda.. The Isoniazid encapsu1ation mo liposomes varied from 4 to 15%, with a gradual release of 24 and 1 hour, when prepared by more crystalline or amorphous microstructures, respectively. The lipid particles produced by spray drying presented a controlled release of drugs, being useful as powders for pulmonary administration or pro-liposomes. Results obtained in pilot scale showed the feasibility for the scaling up of the process, through the production of particles with similar properties to the ones produced by laboratory scale
