Estudo do efeito do naproxeno na regeneração da nadadeira caudal do peixe teleosteo Cyprinus carpio (carpa)

RESUMO

The fins of teleosts show a very fast capacity for regeneration, what make them one important and appropriate biologic model for study of the drugs that can affect their development. The rays of fins are formed, for the most part, of collagen and glycosaminoglycans and, as showed on literature reports, some antiinflammatory drugs, steroidal and nonsteroidal, may affect the extracellular matrix composition of connective issue. In the present investigation we studied the effect of naproxen, a non-steroidal antiinflammatory drug used for the treatment of rheumatic disease, during the regeneration process of the tail fin of the teleost fish Cyprinus carpio (carp). The beginning of the experiment was conducted using 2 aquaria containing 6 liters of water each were set up. The water was clear and decWorinated, the temperature was kept at 24°C and under constant aeration. The naproxen was dissolved in one of them to obtain the dose of 10.4 mg/L, while the other contained on1y water and was used as a control. Forty-two alevins were anesthetized with benzocaine (1 : 1 0000) and their tail fins were transversely amputated. After amputation, the fishes were divided into 2 groups of 21 animaIs each. One group was placed in the aquarium containing the drug and the other was placed in the control aquarium. The fishes were left in the aquaria until the occurrence of regeneration. Half of the water in each aquarium was exchanged daily with clean water until the end of the experiment and half of the dose of naproxen, 5.2 mg/L, was added to the experimental aquarium in order to restaured the initial concentration. The animaIs were anesthetized and sacrificed and the regenerating fins were excised and fixed at intervals of 1,2,4,6,8, 10 and 12 days after amputation. Three specimens per time point were used. The collected samples were processed for standard and polarized light microscopy and for transmission electron microscopy. In general, naproxen at the dose used in the present study did not affect the organization of the extracellular matrix components and the mineralization of the fundamental substance of the lepidotrichia during the process of tail fin regeneration. Since the effect of naproxen, as well as of other non-steroidal anti-inflammatory drugs, depends on the dose used, the route of administration and the metabolism of the animal in which the drug is being tested, higher doses of the drug may perhaps delay or even fully inhibit this process, possibly also provoke disorganization of the lepidotrichial matrix
