RESUMO
In this work, the effects of biocatalysts preparation conditions on the behavior of ethanol oxidation process to acetic acid were studied and a cellular inoculum preparation method was developed. Experiments involving free and immobilized Acetobacter cells in hydrophilic gels containing k-carrageenan, sodium alginate or citric pectin on their composition were carried out in shaking flasks and in a non-conventional three-phase fluidized bed reactor. The porous structure of gels was evaluate by the permeability of beds composed of gel particles. The kind of support and the matrix preparation conditions affect the biocatalyst activity and stability. Due to physical, chemical and biological factors, the variables involved in the immobilization process simultaneously affect the success of the acetic fermentation. The choice of the immobilization conditions should take into account the bacteria (being too small and having low salinity resistance), the aeration demanded (too large), the reaction product (too acid) and the volatile substrate (ethanol). The tests carried out on the bioreactor showed a good mechanical stability of biocatalyst and need of an ethanol evaporation efficient control, due to high aeration levels in the system
