Estudo da atividade das fosfoesterases em tecido de granulação sob efeito de baixas doses de radiação de eletrons

RESUMO

This work had the purpose to seek the effect of low doses of electron radiation on the activity of phosphoesterases in granulation tissue, as this enzymatic group has a major role in the new tissue formation. With that purpose, the animais underwent surgical procedure, receiving a subcutaneous insertion of a disc of PVC foam, averaging 8 milimeters diameter and 4 milimeters thick, at a distance of 3 centimeters away from the incision. This procedure was repeated on eighty-four animais, which were divided into two experimental groups, the irradiated group and the contrai group, each one with forty-two animais. In their tum, the groups had their animal distributed into seven development times for the granulation tissue: five, 7, 10, 14, 17, 20, and 24 days, performing groups of six animais. A linear accelerator was used for irradiation, with a 6 MeV energy electron beam, at a distance of 100 centimeters frem the source to foam, and with a simple dose of 1.0 Gy , 1 centimeter deep. Irradiation was performed three days afier surgical procedure, with the body protected by a lead cover, four milimeters thick, with an aperture allowing an exposition of five milimeters surrounding the foam diameter. Afier the proposed times, the formed tissues were removed and the specific ativity of three enzymes was measured: - The Adenosine Triphosphatase (ATPase) showed a decreased activity, when compared to control, seventeen days afier irradiation. - The Alkaline Phosphatase showed, at eleven days, an increased activity in the irradiated group, decreasing at fourteen and twenty-one days but not significant when compared to the control group. - The 5 Nucleotide Phosphodiesterase showed an increased activity in the irrradiated animais frem two to eleven days, with significant difference from the control groups. At fourteen days a decreasing activity occurred, statistically significant, when compared to the non-irradiated group, followed by a new increasing from seventeen to twenty-one days in the animais belonging to the irradiated group with statistically significant difference at twenty-Oone days, when compared to the control group
