Efeito diabetogenico da aloxana em ratos submetidos a desnutrição proteica

RESUMO

In the present study we examined the effect of protein dietary levei on pancreatic function evaluated by measurements, of serum glucose levels in the fed state, serum glucose and insulin levels during an oral glucose tolerance test, and serum glucose levels during a subcutaneous insulin tolerance test performed after administration of a single endovenous, dose or alloxan to rats fed a low (6%) or a normo (17%) protein. In a separate set of experiments, we examined insulin release in response to glucose and arginine and glucose oxidation rates in pancreatic islets treated "in vitro" with alloxan, isolated from rats fed the low and the normo protein diets. After alloxan administration the incidence of mild hyperglycemia in the fed state (diabetes) was higher among rats fed normo protein diet than among those fed low protein diet. After glucose load the area under serum glucose cUlve (AG) was lower in low protein rats than in normal proteins rats, while then were no differences between the two groups in the area under serum insulin curve (AI). In addition, after an insulin load, the serum glucose disappearance rate (KITT) was higher in lower protein than in normal protein rats. Alloxan induced significant decrease in glucose oxidation and in glucose induced and arginine induced insulin release by islets isolated from normal and low protein rats. The decrease in both insulin release and glucose oxidation was greater in normo than in low protein rat islets. In summary, alloxan was less effective in producing hyperglycemia (diabetes) in rats fed low protein diet than normo protein diet. This protection was associated to an increased peripheral sensibility to insulin action and a better préservation of glucose oxidation and glucose induced insulin release by pancreatic islets exposed to alloxan in protein restricted than in normal protein rats
