RESUMO
Human granulocyte colony-stimulating factor (hG-CSF) has been used as a hematopoietic growth factor due to its efficacy against neutropenia and chemotherapy-induced leucopenia. This project was approved by the Ethics Committee for Animal Use (CEUA-UECE, n 12/2005) and the National Technical Commission on Biosafety (CTNBIO, CQB n 0228/2006). Our group obtained a male and a female goat transgenic for hG-CSF. The two transgenic animals and non-transgenic were monitored up to 285 days-of-age. On 45 days and biweekly thereafter blood samples were collected for hematological evaluation (total and differential white blood cell counts), serum biochemistry (urea, creatinine, glucose, AST and ALT) and hG-CSF analysis. Transgenic goats presented the leucocyte count 2.5 to 3.7-fold greater than non-transgenics. This leukocytosis have a strong relationship with neutrophilia (R2=0.97), which was followed by low (<20pg/mL) or null serum levels of hG-CSF. Strikingly, although the neutrophil counts decreased in transgenic goats before they reached 157 days-old, the neutrophilia persisted until 285 days-of-age. No changes in other cell counts were detected. The serum biochemistry profiles indicated normal liver and renal functions and the weight gain points to the typical body development. In conclusion, hG-CSF-transgenic young goat presented persistent neutrophilia, with low or absent ectopic citokyne expression, and remained without general health disorders
